| HGNC Family | Non-coding RNAs |
| Name | microRNA 3140 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In this context, circCCDC66 acts as a molecular sponge for miR‑3140, sequestering it and consequently relieving its suppressive effects on autophagy. This regulatory mechanism facilitates the progression of hypoxia‐exposed colorectal cancer cells, suggesting that modulating miR‑3140 activity could represent a novel therapeutic strategy."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Disparate from the role of miR‑3140 in cancer progression, structural investigations into the tweety homolog (TTYH) family elucidated a unique fold and Ca2+‑dependent quaternary organization of TTYH2 and TTYH3. These studies, while not addressing miR‑3140, provide insight into Ca2+‑regulated protein interactions, membrane‐spanning dimers, and potential roles in cell adhesion and developmental signaling. Such mechanistic insights into TTYH structure contribute to our growing understanding of the diverse molecular pathways that converge on processes like cellular migration."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "Collectively, these findings underscore the multifaceted nature of cellular regulation in both cancer and normal physiology. While miR‑3140’s suppression of autophagy is a critical factor in hypoxic colorectal cancer cell survival and invasiveness, complementary studies on membrane proteins such as TTYHs expand our overall grasp of calcium‐mediated cellular mechanisms that could intersect with miRNA regulatory networks. Ongoing research integrating these diverse molecular insights promises to enhance future therapeutic strategies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jin Feng, Zhong Li, Ling Li, et al.
