| HGNC Family | Non-coding RNAs |
| Name | microRNA 3148 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐3148 has emerged as an important post‐transcriptional regulator in immune function by targeting Toll-like receptor 7 (TLR7). Specifically, the non-risk allele of TLR7 contains a binding site for miR-3148, which facilitates transcript degradation and limits TLR7 expression. In contrast, the risk-associated allele shows reduced binding and slower degradation of TLR7 mRNA, leading to increased protein levels and a heightened risk for systemic lupus erythematosus."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its immunoregulatory role, miR-3148 has been implicated in the pathogenesis of chronic thromboembolic pulmonary hypertension (CTEPH). Analyses of CTEPH samples revealed that miR-3148 is significantly downregulated compared with controls, and it regulates a large network of target genes. Notably, some of these targets, such as the androgen receptor (AR) and Protein Kinase C Alpha (PRKCA), are enriched in key signaling pathways associated with cancer, glioma, and ErbB signaling, suggesting that miR-3148 may influence disease progression through modulation of these pathways."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFurthermore, in the context of cervical cancer, miR-3148 functions as a tumor suppressor. Its expression is downregulated in cervical cancer cells, and its upregulation leads to the inhibition of cell proliferation and induction of apoptosis. At the molecular level, bromodomain containing 7 (BRD7) enhances miR-3148 transcription, and miR-3148 directly targets the 3’ untranslated region of Wnt3a mRNA, thereby suppressing the Wnt3a/β-catenin signaling pathway. This dual regulatory mechanism underscores the therapeutic potential of miR-3148 in mitigating tumor growth and progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yun Deng, Jian Zhao, Daisuke Sakurai, et al.
