| HGNC Family | Non-coding RNAs |
| Name | microRNA 3163 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‑3163 has emerged as a key post‑transcriptional regulator in cancer by directly binding to specific target mRNAs. In non‑small cell lung cancer, miR‑3163 targets the 3′‑UTR of Skp2 mRNA, thereby inhibiting its translation and leading to reduced Skp2 protein levels with a subsequent increase in the cell cycle inhibitor p27. This effect is further enhanced by the long non‑coding RNA Meg3, which appears to coordinate with miR‑3163 to suppress Skp2 expression and inhibit tumor growth."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn hepatocellular carcinoma, miR‑3163 contributes to tumor suppression by targeting ADAM‑17, a metalloprotease that activates the Notch signaling pathway; its inhibition results in enhanced sensitivity of cancer cells to targeted agents like sorafenib. In pancreatic cancer, the lncRNA DGCR5 sequesters miR‑3163, resulting in the upregulation of DNA topoisomerase 2‑α (TOP2A) and activation of the Wnt/β‑catenin pathway, whereas restoring miR‑3163 activity suppresses these oncogenic signals and improves chemotherapeutic response."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFurthermore, in cervical cancer and nasopharyngeal carcinoma, miR‑3163 plays a crucial tumor‑suppressive role. In cervical cancer, diminished miR‑3163 expression correlates with increased levels of LASP1 and is associated with poor clinical outcomes, a situation exacerbated by the aberrant sponging activity of lncRNA CBR3‑AS1. Similarly, in nasopharyngeal carcinoma, miR‑3163 is competitively inhibited by lncRNA CRNDE, leading to elevated expression of the pro‑metastatic transcription factor TWIST1 and enhanced tumor progression.
