MIR3168 Gene

HGNC Family Non-coding RNAs
Name microRNA 3168
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Elevated levels of miR-3168 have been observed in HCC tissues, where its expression correlates with poor patient prognosis. Functional assays demonstrate that miR-3168 enhances HCC cell proliferation, promotes liver cancer stem cell self-renewal, and increases tumorigenicity. Mechanistically, miR-3168 directly targets the 3′ untranslated region of p53 mRNA, thereby downregulating this crucial tumor suppressor and impairing its role in cell cycle regulation and apoptosis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nRescue experiments show that knockdown of p53 reverses the differences in cell proliferation, self-renewal, and tumorigenic potential between miR-3168-deficient and control HCC cells, underscoring the centrality of the miR-3168–p53 axis in cancer progression. Additionally, higher miR-3168 expression is associated with resistance to cisplatin-based therapy, suggesting that miR-3168 not only drives malignant behavior but may also serve as a valuable biomarker for predicting chemoresistance in HCC patients."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Tingting Lu, Tao Han, Mingfang Zhao "}, {"type": "b", "children": [{"type": "t", "text": "miR-3168 promotes hepatocellular carcinoma progression via downregulating p53."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Carcinogenesis (2022)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1093/carcin/bgac062"}], "href": "https://doi.org/10.1093/carcin/bgac062"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "35926447"}], "href": "https://pubmed.ncbi.nlm.nih.gov/35926447"}]}]}]}
