MIR3180-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 3180-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Mechanistic studies revealed that lncKRT16P6 directly binds to MIR3180-1 (functionally equivalent to miR‑13180), effectively “sponging” it. Under normal conditions, MIR3180-1 exerts tumor‐suppressive effects by inhibiting the expression of GATAD2A, a gene that when upregulated promotes malignant traits."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Disruption of this balance was evidenced by the finding that inhibiting lncKRT16P6 expression restored MIR3180-1 activity, thereby reducing TSCC cell migration, invasion and proliferation through the downregulation of GATAD2A. Conversely, direct inhibition of MIR3180-1 reversed these beneficial effects, leading to enhanced tumorigenic properties. This clearly implicates MIR3180-1 as a key mediator that, when not sequestered by lncKRT16P6, helps restrain TSCC malignancy."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "These insights into the ceRNA regulatory pathway highlight the potential of MIR3180-1 not only as a biomarker for assessing TSCC prognosis but also as a promising therapeutic target.
