MIR3194 Gene

HGNC Family Non-coding RNAs
Name microRNA 3194
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have uncovered that miR-3194 plays a multifaceted role in modulating cancer stem cell phenotypes, notably in the context of human papillomavirus (HPV)‐associated oropharyngeal squamous cell carcinoma (OPSCC). In HPV16-positive cells, miR-3194-5p is upregulated in the migratory (CD44high/EpCAMlow) cancer stem cell population, and its aberrant expression correlates with increased metastatic potential. These findings suggest that miR-3194-5p may serve as a promising biomarker and therapeutic target to block metastasis in HPV-driven malignancies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nIn esophageal cancer, miR-3194-3p has been identified as an integral component of a competing endogenous RNA network involving the long noncoding RNA KCNMB2-AS1. Here, KCNMB2-AS1 sequesters miR-3194-3p, thereby relieving miR-3194-3p-mediated repression of its target gene PYGL. The upregulation of PYGL in this axis contributes to enhanced cell proliferation, invasion, migration, and maintenance of stemness features, implicating miR-3194-3p as a key regulator in esophageal tumorigenesis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nAdditionally, miR-3194-5p has been implicated in the modulation of vascular endothelial cell function under hypoxic conditions, a critical component of coronary heart disease pathology. In hypoxia-exposed human umbilical vein endothelial cells (HUVECs), LINC00926 is highly expressed and acts by binding miR-3194-5p. This interaction disrupts the suppression of JAK1, leading to enhanced JAK1/STAT3 signaling, which in turn impairs cell proliferation, migration, tube formation, and promotes apoptosis. Such findings not only underscore the role of miR-3194-5p in endothelial dysfunction but also provide insights into its potential as a therapeutic target in vascular disease"}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Martin Hufbauer, Margaret Maltseva, Jeannine Meinrath, et al.
