| HGNC Family | Non-coding RNAs |
| Name | microRNA 323a |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nStudies across multiple cancers have revealed that MIR323A—often examined as its mature forms miR‑323a‑3p and miR‑323‑5p—plays diverse roles in modulating tumor cell behavior. In glioblastoma and other brain tumors, MIR323A expression has been linked with reduced proliferation and migration, with some reports implicating its targeting of signaling components (for example, elements of the TGF‑β cascade) to improve survival outcomes. In pancreatic ductal adenocarcinoma and osteosarcoma, restoration of MIR323A levels suppresses key oncogenic processes, including cellular glycolysis via targets such as hexokinase‑2 or lactate dehydrogenase A. Similarly, in bladder cancer and neuroblastoma, overexpression of MIR323A limits epithelial–mesenchymal transition and cell motility by directly downregulating receptor tyrosine kinases (such as MET, c‑Met, ErbB3, EGFR) or cell cycle regulators like cyclin D1. Conversely, in non‑small‑cell lung cancer, elevated MIR323A expression appears to promote oncogenic signaling via inhibition of tumor suppressive targets like TMEFF2 and subsequent activation of the AKT/ERK pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of reproductive and neurological disorders, MIR323A has also been implicated in the regulation of cell proliferation, apoptosis, and differentiation. In polycystic ovary syndrome (PCOS), divergent observations have emerged: one study reported that exosomal MIR323A elevation in adipose‑derived mesenchymal stem cells promotes cumulus cell proliferation and suppresses apoptosis via targeting PDCD4, alleviating PCOS features, whereas another study observed downregulation of MIR323A in human cumulus cells with subsequent effects on steroidogenesis mediated by direct suppression of IGF‑1. In major depressive disorder, increased MIR323A levels in key brain regions correlate with altered expression of ERBB4—a neuregulin receptor—and modulate emotional behaviors in animal models. Furthermore, a genetic variant within the MIR323B coding region associates with an elevated risk of recurrent pregnancy loss through upregulation of mature MIR323B and consequent downregulation of its target, paired‑box 8; MIR323A levels have even been proposed as biomarkers for cardiomyopathy in Friedreich’s ataxia."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional investigations extend the functional spectrum of MIR323A to other pathologic settings. In cerebral infarction models, overexpression of MIR323 results in increased neuronal toxicity and diminished cell growth via direct targeting of SMAD3 in the TGF‑β1 pathway, while in glioma U373 cells, enforced MIR323‑5p expression enhances apoptosis through downregulation of the IGF‑1 receptor.
