| HGNC Family | Non-coding RNAs |
| Name | microRNA 324 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In several studies, upregulation of miR‐324 (for example, via metformin treatment in breast cancer) has been shown to promote cell death mechanisms such as ferroptosis by downregulating targets like GPX4, while in other tumor types miR‐324 functions as a tumor suppressor by inhibiting cell proliferation, migration and invasion. In hepatocellular carcinoma, ectopic expression of miR‐324‐5p suppresses invasive behavior through direct targeting of ETS1 and SP1; in colorectal cancer it limits cell viability and invasive capacity by downregulating ELAVL1; and in nasopharyngeal and thyroid carcinomas, as well as in cholangiocarcinoma, aberrant levels of miR‐324 are associated with therapy resistance and poor prognosis. These findings collectively indicate that the precise control of miR‐324 expression and the selection of its arm play key roles in modulating oncogenic signaling pathways and the tumor microenvironment."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond malignancy, MIR324‐5p has been implicated in regulating non‐cancer processes that include host defense and the maintenance of tissue homeostasis. In models of viral infection, miR‐324‐5p represses pathogen replication—notably by targeting components of the influenza viral genome as well as cellular inhibitors of interferon responses—thereby contributing to antiviral defense. In cardiovascular and metabolic contexts, miR‐324‐5p protects endothelial progenitor cells from oxidative stress, modulates cardiomyocyte survival during hypoxia/reoxygenation through regulation of SOCS3 and TNF/NF‐κB signaling, and influences glucose and lipid metabolism via targeting of key regulators such as ROCK1. Several studies have demonstrated that long non‐coding RNAs and circular RNAs act as “sponges” for miR‐324, thereby modulating its availability and downstream target regulation. For example, in neurodegenerative models the lncRNA RP11–543N12.1 sequesters miR‐324‐3p to affect neuronal apoptosis, while in reproductive and endocrine disorders such as polycystic ovary syndrome, specific circRNAs bind miR‐324‐3p to regulate kinases like MAP2K6.
