MIR33A Gene

HGNC Family Non-coding RNAs
Name microRNA 33a
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR33A is an intronic microRNA encoded within the SREBP‐2 gene that plays a pivotal role in maintaining lipid homeostasis. It directly represses key mRNAs—including those coding for the ATP‐binding cassette transporters ABCA1 and ABCG1—to reduce cholesterol efflux and regulate high‐density lipoprotein (HDL) synthesis. In addition, MIR33A influences fatty acid oxidation and insulin signaling by targeting enzymes and signaling intermediates, thereby integrating cholesterol biosynthesis with lipid degradation. Passenger forms of MIR33 (miR‐33*) further contribute to this extensive regulatory network, with alterations in MIR33A expression linked to atherosclerosis, non‐alcoholic fatty liver disease, and familial hypercholesterolemia."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn various neoplastic settings, MIR33A exerts multifaceted effects on tumor cell proliferation, cell cycle progression, and metastatic behavior. In some contexts it functions as a tumor suppressor—by targeting oncogenic regulators such as Pim-1, beta‐catenin, and PBX3, it limits cell growth, invasion, and the self‐renewal of cancer stem–like populations. Conversely, aberrant expression of MIR33A or its sequestration by competing endogenous RNAs has been associated with enhanced malignant potential in diverse cancers including glioma, melanoma, breast cancer, and non–small cell lung cancer, underscoring its context–dependent influence on tumor biology."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, MIR33A is increasingly recognized for its roles beyond lipid metabolism and cancer. In hepatic stellate cells, for instance, MIR33A modulates TGF–β–induced fibrogenic activation, contributing to liver fibrosis. Altered circulating levels of MIR33A have been associated with cardiovascular pathologies such as coronary artery disease, while dysregulation in placental tissues has been linked to gestational diabetes mellitus. In the musculoskeletal system, interactions between long noncoding RNAs and MIR33A can affect osteogenic differentiation and may hold therapeutic promise for osteoporosis. These diverse functions illustrate the potential of targeting MIR33A as a novel therapeutic strategy for a broad spectrum of metabolic and degenerative disorders."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "22", "end_ref": "26"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "S Hani Najafi-Shoushtari, Fjoralba Kristo, Yingxia Li, et al.
