| HGNC Family | Non-coding RNAs |
| Name | microRNA 34a |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. This miRNA is a member of the highly conserved miR-34 family. This miRNA functions as a tumor suppressor and dysregulation or loss of the host gene from which this miRNA is processed is associated with cancer progression in numerous cell types. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR-34a is a pivotal tumor‐suppressive microRNA whose expression is directly induced by p53 following cellular stress, DNA damage, or oncogene activation. Its transcriptional upregulation initiates a feed‐forward loop that reinforces p53 activity and leads to cell cycle arrest, apoptosis, and senescence through the repression of key regulators—including SIRT1, cyclin D1, CDK6, E2F family members, and pro‐apoptotic targets such as p21 and PUMA. In multiple cancer types, silencing of miR-34a through genomic deletions (often at 1p36) or promoter hypermethylation interrupts these tumor‐suppressive pathways, thereby contributing to unchecked proliferation and resistance to chemotherapeutic agents."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its classical functions in cell cycle control, miR-34a critically modulates pathways governing epithelial-to-mesenchymal transition (EMT), metastasis, and cancer stem cell fate. By directly targeting pivotal inducers of EMT and stemness—such as SNAIL, SLUG, c-Met, Notch1, and key components of the Wnt/β-catenin cascade—it impairs tumor cell migration, invasion, and self-renewal. Reduced miR-34a levels in tumors are associated not only with enhanced metastatic potential in cancers like neuroblastoma, glioma, and non–small cell lung carcinoma but also with immune evasion via modulation of PD-L1 and inflammatory cytokine signaling. Moreover, miR-34a has been implicated in governing cell-fate decisions in cancer stem cells and in promoting differentiation in diverse cell types."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "32"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nThe clinical relevance of miR-34a is underscored by its emerging role as a prognostic biomarker and a therapeutic target. In several cancers—including neuroblastoma, prostate cancer, pancreatic cancer, chronic lymphocytic leukemia, and non–small cell lung cancer—diminished miR-34a expression correlates with aggressive disease, poor treatment response, and drug resistance. Preclinical models demonstrate that restoration of miR-34a, either through direct replacement or via combination with other tumor suppressive microRNAs (such as let-7), can inhibit tumor growth, overcome chemoresistance, modify the tumor immune microenvironment, and even regulate metabolic derangements seen in conditions like non-alcoholic fatty liver disease.
