MIR3622A Gene

HGNC Family Non-coding RNAs
Name microRNA 3622a
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Functional studies demonstrated that restoration of miR-3622a-3p levels suppresses proliferation, invasion, and stemness properties by directly targeting and degrading SALL4 mRNA, thereby inhibiting epithelial–mesenchymal transition and promoting apoptosis. These effects were confirmed in various in vitro assays as well as in vivo tumor xenograft and organoid models, underscoring its tumor‐suppressive role in colorectal cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Conversely, in bladder cancer, miR-3622a acts as an oncogenic microRNA. Here, miR-3622a expression is negatively correlated with overall patient survival and functions to potentiate malignant behavior by downregulating the metastasis suppressor gene LASS2. Overexpression of miR-3622a in bladder cancer cells leads to increased proliferation and invasion, highlighting its role in promoting tumor progression in this context."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "Collectively, these studies reveal that the function of miR-3622a is highly context-dependent: while it serves as a tumor suppressor in colorectal cancer via SALL4 inhibition, it concurrently acts as a tumor promoter in bladder cancer through attenuation of LASS2. These insights into the dualistic nature of miR-3622a underscore the complexity of microRNA regulation in cancer and may provide novel avenues for tailored therapeutic strategies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Shuchen Chang, Guangli Sun, Dan Zhang, et al.
