| HGNC Family | Non-coding RNAs |
| Name | microRNA 363 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

For example, during human placental trophoblast differentiation the miR‐106a∼363 cluster is downregulated in concert with the induction of key differentiation markers, and aberrant regulation of these miRNAs disrupts syncytiotrophoblast formation. In adipose‐derived stromal cells, overexpression of miR‐363 markedly impairs mitotic clonal expansion by targeting the transcription factor E2F3, thereby blunting terminal adipocyte differentiation. Moreover, in the bone marrow microenvironment miR‐363–5p regulates the expression of angiocrine factors that modulate endothelial tube formation and hematopoietic precursor adhesion; additional studies even implicate miR‐363 in the specification of cardiomyocyte subtypes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn numerous malignancies the predominant function of miR‐363–3p (and in several settings, miR‐363–5p) is tumor suppressive through direct targeting of key oncogenic mediators. In hepatocellular, renal, colorectal, endometrial and lung cancers, miR‐363–3p suppresses oncogenic signaling by directly reducing expression of factors such as USP28, MCL‐1, S1PR1, CREB1, NOTCH1, PCNA, SP1, SphK2 and HMGA2—actions that collectively block cell proliferation, migration, invasion and chemoresistance. In colon cancer, miR‐363 targeting of GATA6 mediates downstream repression of REG4, while in breast and cervical tumors, diminished exosomal miR‐363–5p is emerging as a promising biomarker for lymph node metastasis and poor prognosis. In addition, miR‐363 levels and activity are frequently modulated by a network of long noncoding RNAs (for example, MALAT1, FOXD2‐AS1, NNT‐AS1, OIP5‐AS1, HNF1A‐AS1 and NORAD) that act as endogenous sponges to fine tune its availability and, consequently, its tumor‐suppressive outputs."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "30"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nInterestingly, miR‐363 can also exhibit oncogenic (oncomiR) properties in a context‐dependent manner. Similar pro‐oncogenic roles have been observed in subsets of head and neck cancers associated with human papillomavirus, in glioma where miR‐363 overexpression correlates with higher tumor grade and enhanced cell survival, and in T‑cell acute lymphoblastic leukemia where miR‐363–3p supports proliferation by repressing PTEN and BIM.
