| HGNC Family | Non-coding RNAs |
| Name | microRNA 3646 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In patients with acute coronary syndrome, miR‑3646 is markedly upregulated compared with healthy controls, and its expression level correlates with disease severity. Experimental models demonstrated that silencing miR‑3646 alleviates the inflammatory response associated with acute coronary events, indicating that this miRNA may serve as both a valuable diagnostic biomarker and a potential therapeutic target in these conditions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nDysregulation of miR‑3646 has also been reported in lung adenocarcinoma. In this setting, elevated miR‑3646 levels were found to promote malignant cellular behaviors by enhancing cell proliferation and adhesion while reducing apoptosis. Mechanistic investigations revealed that miR‑3646 directly targets the gene SORBS1, leading to disruption of the JNK signaling pathway—a process that appears to drive tumor progression. These findings suggest that miR‑3646 contributes to lung cancer pathogenesis and may offer a novel target for therapeutic intervention."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its roles in acute coronary syndrome and lung adenocarcinoma, miR‑3646 has been implicated in modulating the inflammatory response of vascular smooth muscle cells. Studies on angiotensin II–induced human vascular smooth muscle cells indicate that upregulation of miR‑3646 is associated with increased inflammatory signaling, which may contribute to the development and progression of hypertension and coronary artery disease.
