| Name | microRNA 3648-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Functional studies using the prostate cancer cell line LNCaP demonstrated that enhanced expression of miR-3648 leads to increased cell viability, colony formation, and anchorage-independent growth, while silencing miR-3648 produces the opposite effect. These findings indicate that miR-3648 acts as an oncomiR in the context of prostate cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistic investigations identified the tumor suppressor APC2—a negative regulator of the Wnt/β-catenin pathway—as a direct target of miR-3648. Using luciferase reporter assays, researchers confirmed that miR-3648 binds to the 3' untranslated region of APC2 mRNA, suggesting that miR-3648 promotes tumorigenesis by downregulating APC2 expression. This interaction leads to aberrant activation of the Wnt/β-catenin signaling pathway."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nConsequently, the activation of the Wnt/β-catenin pathway results in altered expression of key regulators of the cell cycle, notably through upregulation of cyclin D1 and cyclin E1 and downregulation of the cell cycle inhibitor p21. These molecular changes facilitate the transition from the G1 phase to the S phase, thereby accelerating cell proliferation. A notable negative correlation between miR-3648 and APC2 expression in prostate cancer tissues further underscores its potential role in disease progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Rongwei Xing "}, {"type": "b", "children": [{"type": "t", "text": "miR-3648 Promotes Prostate Cancer Cell Proliferation by Inhibiting Adenomatous Polyposis Coli 2."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "J Nanosci Nanotechnol (2019)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1166/jnn.2019.16413"}], "href": "https://doi.org/10.1166/jnn.2019.16413"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "31196256"}], "href": "https://pubmed.ncbi.nlm.nih.gov/31196256"}]}]}]}
