MIR369 Gene

HGNC Family Non-coding RNAs
Name microRNA 369
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent evidence demonstrates that both strands of the miR-369 duplex function largely as tumor suppressors across a spectrum of malignancies. In familial breast and ovarian cancer patients, no allelic variants were identified in hsa‐miR‑369, suggesting that its integrity is maintained despite cancer predisposition (1). In glioblastoma and papillary thyroid cancer, downregulation of miR‑369‑3p is associated with increased oncogenic signaling, with its restoration suppressing proliferation and migration by directly targeting mediators such as components of neuregulin pathways and TSPAN13 (6, 8). In gastric cancer, concomitant inactivation of both miR‑369‑5p and miR‑369‑3p due to promoter methylation leads to dysregulation of key effectors including JUN and AKT, while in hepatocellular carcinoma distinct targets—including SOX4, ACTN4, and HOXA13—mediate its antiproliferative and pro‐apoptotic effects. Similarly, in prostate cancer the sponging of miR‑369‑3p by lncRNA SOX2‑OT results in upregulation of CFL2 and promotes tumor progression (9, 10, 12, 13, 14, 15)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its oncogenic roles, miR‑369 is critically involved in developmental and neurological contexts. In mesenchymal stromal cells, up‐regulation of miR‑369‑5p during long‐term culture impairs adipogenic differentiation—at least partly via direct targeting of genes like FABP4—and suggests a role in cellular senescence (2). In the nervous system, altered binding of hsa‑miR‑369‑3p to its target mRNAs may influence Alzheimer’s disease risk (3), while in embryonic stem cells, epigenetically regulated miR‑369 expression stabilizes translation of splicing factors that modulate metabolic enzyme isoforms, thereby contributing to cell reprogramming (5). Finally, although a connection with Hirschsprung disease remains to be fully elucidated, altered miR‑369‑3p levels may also be relevant in neurodevelopmental disorders (7)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the realm of inflammatory and immune-mediated conditions, miR‑369‑3p emerges as an important regulator of immune signaling. In lung transplant recipients, decreased levels of miR‑369‑5p in mononuclear cells correlate with deregulation of TGF‑β and B cell receptor pathways, contributing to chronic rejection manifested as bronchiolitis obliterans syndrome (4). Furthermore, miR‑369‑3p is enriched in extracellular vesicles released by cancer-associated fibroblasts in lung squamous cell carcinoma, where it modulates NF1 expression and downstream MAPK signaling (11). Notably, in inflammatory bowel disease, miR‑369‑3p directly targets PDE4B, leading to increased cellular cAMP levels and activation of anti-inflammatory signaling cascades; additional immunomodulatory effects of miR‑369‑3p have been linked to its influence on the immunoproteasome (16, 17)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Chiara Pastrello, Jerry Polesel, Lara Della Puppa, et al.
