| HGNC Family | Non-coding RNAs |
| Name | microRNA 374b |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‑374b (and its mature form miR‑374b‑5p) has emerged as an important regulator in cancer biology. In a number of malignancies—including triple‐negative breast cancer, gastric cancer, colorectal cancer, T‑cell lymphoblastic lymphoma, pancreatic cancer, colon cancer, cervical cancer, ovarian cancer, hepatocellular carcinoma, gastrointestinal stromal tumors, prostate cancer, non‑small cell lung cancer, and breast cancer—aberrant expression of miR‑374b has been observed. In many of these tumors the transcript is downregulated, and its restoration leads to inhibition of cell proliferation, invasion, metastasis and even resistance to chemotherapy. In these contexts, miR‑374b appears to function as a tumor suppressor by targeting oncogenic drivers such as LRH‑1, FOXP1, cyclin D1 (CCND1), transforming growth factor alpha (TGFA), JAM‑2, AKT1 and related signaling cascades including the p53 pathway. In contrast, in certain settings such as hepatocellular carcinoma and gastrointestinal stromal tumors, miR‑374b is upregulated and promotes cell viability by modulating the PI3K/Akt pathway through suppression of PTEN and influencing SMG1‐dependent mechanisms."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "14"}]}, {"type": "t", "text": "\n\nMorover, studies in non‐neoplastic conditions have revealed that miR‑374b plays pivotal roles in regulating cell survival and inflammatory responses. In neurodegenerative and neuropsychiatric disorders such as amyotrophic lateral sclerosis and major depressive disorder, altered miR‑374b levels have been detected—with lower levels correlating with disease activity and treatment response. Similarly, in conditions like multiple sclerosis and inflammatory bowel disease, miR‑374b is upregulated and appears to contribute to the orchestration of pro‑inflammatory signaling. Its increased expression in oral mucosal cells following inflammatory or tobacco‑related insults further underscores its involvement in immune regulation and tissue stress responses."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "19"}]}, {"type": "t", "text": "\n\nBeyond oncology and immune‐mediated disorders, miR‑374b has relevance in bone metabolism and reproductive health. Elevated circulating levels of miR‑374b‑5p are associated with postmenopausal osteoporosis, where it functions as a critical suppressor of osteoblast differentiation by directly targeting key regulators of the Wnt signaling pathway, including Wnt3 and Runx2. In the reproductive arena, decreased miR‑374b levels in seminal plasma correlate with increased sperm DNA fragmentation, suggesting its potential as an adjunct biomarker for idiopathic male infertility. Additionally, emerging data implicate miR‑374b in the pathogenesis of osteoarthritis, further broadening its spectrum of biological influence."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "20", "end_ref": "22"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Luciano Cascione, Pierluigi Gasparini, Francesca Lovat, et al.
