MIR374C Gene

HGNC Family Non-coding RNAs
Name microRNA 374c
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have highlighted the critical role of miR-374c-5p as a tumor suppressor in diverse cancers. In breast cancer, miR-374c-5p is expressed at low levels, and its upregulation has been shown to significantly inhibit cell proliferation, migration, and epithelial–mesenchymal transition (EMT), while promoting apoptosis. Mechanistically, miR-374c-5p interacts with and downregulates TAF7, which in turn modulates DEPDC1 expression, thereby suppressing tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n \nIn parallel, investigations in hepatocellular carcinoma (HCC) have demonstrated that a similar downregulation of miR-374c-5p is associated with poor patient prognosis and shorter survival. Enhancing its expression in HCC cell lines not only restrains proliferation, migration, and invasion, but also inhibits tumor growth in vivo. This effect is achieved through the direct targeting of the 3′‐untranslated region of PTTG1, which modulates EMT and malignant cell behavior."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n \nCollectively, these findings from breast cancer and HCC studies indicate that miR-374c-5p plays a pivotal role in suppressing tumor development via distinct molecular pathways. Its dual functionality in regulating TAF7/DEPDC1 and PTTG1-mediated transcriptional and posttranscriptional events positions miR-374c-5p as a promising diagnostic marker and therapeutic target in the treatment of malignant tumors.\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Shuai Hao, Wuguo Tian, Yi Chen, et al.
