MIR376A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 376a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn hepatocellular carcinoma (HCC), miR-376a (also referred to as MIR376A2) functions as a tumor suppressor. Its expression is frequently down-regulated in HCC tissues and cell lines, and restoration of its levels inhibits cell proliferation while inducing apoptosis. Mechanistically, miR-376a directly targets p85α (PIK3R1), suggesting that its decreased expression may facilitate oncogenic signaling pathways that contribute to HCC development."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn non‐small cell lung cancer (NSCLC), miR-376a-3p plays a similar suppressive role. Here, a circular RNA, circ-UBE2D2, acts as a molecular sponge for miR-376a-3p, reducing its availability and thereby indirectly promoting the expression of EIF4G2, an important factor in translation initiation. Consequently, diminished miR-376a-3p activity is associated with increased cell proliferation, migration, and invasion, while its overexpression leads to apoptosis. These findings extend the tumor-suppressive functions of miR-376a across cancer types by regulating key pathways involved in cell growth and survival."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yongxia Zheng, Lei Yin, Huan Chen, et al.
