MIR379 Gene

HGNC Family Non-coding RNAs
Name microRNA 379
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAccumulating evidence from multiple studies demonstrates that miR‑379 (and its mature isoform miR‑379‑5p) functions as a potent tumor suppressor in diverse cancer types. In breast, hepatocellular, bladder, cervical, and lung cancers as well as diffuse gliomas and glioma, reduced miR‑379 expression has been associated with increased cell proliferation, migration, invasion, epithelial–mesenchymal transition (EMT) and more aggressive clinical stages. For example, in breast cancer lower miR‑379 expression correlates with elevated Cyclin B1 levels and higher tumor stage, while in hepatocellular carcinoma its restoration inhibits cell migration and invasion by repressing focal adhesion kinase and downstream AKT signaling. Similarly, in bladder and cervical cancers, downregulation of miR‑379 permits increased expression of oncogenic targets that drive disease progression, and in non‑small cell lung cancer (NSCLC) its decreased levels have been linked with chemoresistance and unchecked cell growth through dysregulation of proliferation‐promoting factors."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistic investigations have revealed that miR‑379 exerts its tumor‐suppressive effects by directly targeting multiple pro‑oncogenic mRNAs. In breast cancer, miR‑379 lowers COX‑2 and Cyclin B1 levels, whereas in hepatocellular carcinoma and gastric cancer it targets focal adhesion kinase (FAK) and GRB10 to suppress AKT‐driven signaling pathways. In glioblastoma, sequestration of miR‑379‑5p by the circular RNA circ‑PITX1 de‑represses MAP3K2 expression, promoting tumor progression. Additional mechanisms include direct targeting of CHUK in NSCLC, inhibition of TCF‑4 in laryngeal carcinoma, and suppression of CRKL in cervical cancer. Moreover, miR‑379 activity is modulated by non‑coding RNAs—such as lncRNA LINC00665 and specific circular RNAs—that act as molecular sponges, thereby influencing chemoresistance (for instance, through IGF1 regulation) and other oncogenic processes. Altogether, these findings underscore a multilayered regulatory network in which miR‑379 normally restrains key effectors of cell proliferation, migration, and invasion."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}, {"type": "fg_f", "ref": "12"}, {"type": "fg_fs", "start_ref": "6", "end_ref": "8"}, {"type": "fg_fs", "start_ref": "13", "end_ref": "15"}, {"type": "fg_f", "ref": "11"}, {"type": "fg_fs", "start_ref": "16", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its role in tumor suppression, miR‑379 influences several physiological and disease‐related processes by modulating gene networks involved in metabolism, inflammation, and cellular stress responses. In metabolic contexts, glucocorticoid‑induced miR‑379 affects hepatic lipid handling, and epigenetic studies have linked its expression to key regulators of retinoic acid signaling. In the ovary, aberrant miR‑379‑5p expression impairs granulosa cell proliferation and DNA repair—contributing to premature ovarian insufficiency—and, in polycystic ovarian syndrome (PCOS), androgen‑stimulated exosomal release of miR‑379‑5p has been shown to influence granulosa cell fate and local inflammatory responses. Additionally, altered miR‑379 levels are associated with intestinal barrier dysfunction during ischemia–reperfusion injury, further emphasizing its multifaceted role in regulating cellular homeostasis.
