MIR3913-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 3913-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough none of the provided studies directly address MIR3913‑1, they collectively elucidate the critical role of Roquin proteins—highly conserved RNA‐binding E3 ubiquitin ligases—in maintaining immune homeostasis through post‑transcriptional regulation of key mRNAs. Roquin binds to defined stem–loop structures (e.g. constitutive decay elements and alternative hexaloop motifs) in the 3′‑untranslated regions of transcripts encoding co‑stimulatory receptors (such as ICOS and OX40) and inflammatory mediators, thereby promoting mRNA decay and limiting excessive T‑cell activation. Notably, several reports also describe cooperation with microRNAs (for example, miR‑101 and miR‑146a), which may hint at a broader spectrum of microRNA involvement in this regulatory network—a context that could be relevant when considering the potential, yet unexplored, functions of MIR3913‑1."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": "\n\n"}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistic studies have revealed that Roquin exerts its regulatory effect by localizing to cytoplasmic RNA granules (such as P‑bodies) and recruiting mRNA decay complexes—like the CCR4–NOT deadenylase complex—as well as by interacting with other RNA‒processing factors. The ROQ domain and associated structural modules mediate direct recognition of RNA stem–loops, while additional domains (including the RING and HEPN domains) further modulate interactions and enzymatic activities. Mutations or conditional deletions that impair these functions lead to dysregulated mRNA expression and altered T‑cell differentiation (notably, exaggerated follicular helper and Th17 responses), underscoring the importance of this post‑transcriptional pathway in preventing autoimmunity."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "16"}]}, {"type": "t", "text": "\n\n"}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn vivo models further underscore the biological significance of Roquin‐mediated regulation. Aberrant or diminished Roquin function is associated with a spectrum of pathologies spanning systemic autoimmunity, hyperinflammatory syndromes, and even influences the efficacy of cancer immunotherapies via altered CD8+ T‑cell expansion. In addition, cleavage of Roquin by signaling proteases (for example, MALT1) links T‑cell receptor signaling strength to graded mRNA derepression, thereby fine‑tuning T‑cell differentiation and immune responses. While MIR3913‑1 is not explicitly mentioned in these reports, the extensive interconnection between Roquin function and microRNA pathways suggests that MIR3913‑1—if functionally integrated—could contribute to the nuanced post‑transcriptional control of immune regulators."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "17", "end_ref": "28"}]}, {"type": "t", "text": "\n\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Carola G Vinuesa, Matthew C Cook, Constanza Angelucci, et al.
