| HGNC Family | Non-coding RNAs |
| Name | microRNA 3914-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In one study, candidate miRNAs were screened for their ability to regulate genes involved in this process, and miR-3914—also referenced as MIR3914-2—emerged as a key regulator. This molecule was predicted and later confirmed to modulate the expression of genes critical to extracellular matrix formation and ameloblast differentiation in AM-1 cells, a human ameloblastoma cell line."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "In particular, MIR3914-2 functions by fine-tuning the genetic programs that drive ameloblast differentiation. By suppressing or adjusting the levels of target gene expression through post-transcriptional mechanisms, miR-3914 helps ensure the proper development of the enamel matrix, highlighting its potential role in the pathobiology of amelogenesis imperfecta. This regulatory action suggests that any dysregulation of miR-3914 could contribute to the enamel defects observed in affected individuals."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Overall, the identification of MIR3914-2 as a regulatory miRNA in ameloblast differentiation not only enhances our understanding of the molecular underpinnings of enamel development but also opens promising avenues for future research.
