MIR412 Gene

HGNC Family Non-coding RNAs
Name microRNA 412
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In hemorrhoid tissues, miR‑412-5p expression is markedly decreased relative to normal tissue, a change that relieves its inhibitory control over target mRNAs. Experimental data indicate that miR‑412-5p directly binds to the 3′‑UTR of Xpo1 mRNA, resulting in its silencing and subsequent modulation of downstream signaling. With Xpo1 repressed, the tumor suppressor protein p53 is retained within the nucleus where it is able to promote the expression of cell cycle regulators such as p66SHC and p16. Consequently, the overexpression of miR‑412-5p in endothelial cells leads to reduced proliferation, migration, and vascular structure formation, thereby impeding angiogenesis. In contrast, the downregulation of miR‑412-5p observed in hemorrhoid tissues facilitates increased Xpo1 expression and disrupts p53 nuclear function, ultimately contributing to unchecked vascular endothelial cell division and angiogenesis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Chen Wang, Hong Lu, Chunsheng Luo, et al.
