MIR4289 Gene

HGNC Family Non-coding RNAs
Name microRNA 4289
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In one study, a neuropathic pain model revealed that reduced levels of a microRNA (mmu‑miR‑1449a) in dorsal root ganglion neurons correlated with altered ion channel activity. Specifically, restoration of this microRNA in DRG cells decreased mRNA levels of the TRPA1 and KCNMA1 channel proteins while increasing the transcript for TPTE, suggesting that the microRNA regulates neuronal excitability and may offer a therapeutic route for alleviating neuropathic pain."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a separate investigation related to factors affecting cell proliferation, researchers identified a splice‑variant of TPIP (TPIP‑C2) expressed in human and murine tissues. Over‑expression of this alternatively spliced mRNA in HeLa cells led to marked growth inhibition and caspase‑3‑dependent apoptosis. These findings underscore the regulatory potential of phosphatase‑related transcripts in controlling cell survival and proliferation."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAlthough neither study explicitly mentioned MIR4289, the functional paradigms exemplified by mmu‑miR‑1449a imply that MIR4289 could similarly modulate key regulatory proteins. By potentially targeting transcripts involved in ion channel regulation and phosphatase‑related signaling—as seen with TRPA1, KCNMA1, and TPTE/TPIP pathways—MIR4289 might contribute to reducing aberrant neuronal excitability associated with neuropathic pain and, possibly, to the modulation of cell proliferation. Further research is warranted to define the precise targets and therapeutic potential of MIR4289 in these contexts."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Shan Lu, Sichao Ma, Yunyun Wang, et al.
