MIR4291 Gene

HGNC Family Non-coding RNAs
Name microRNA 4291
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies on imprinted gene clusters have established that long non‐coding RNAs play active roles in cis‐silencing of clusters such as Igf2r/Slc22a2/Slc22a3. For example, investigation of the Air RNA in these regions has revealed that transcription‐mediated chromatin modifications are critical for the monoallelic (imprinted) expression of associated protein–coding genes (see, for example, the work characterizing a 3.7‐kilobase bidirectional silencer and the requirement of full Air RNA expression for silencing). Although MIR4291 is not mentioned explicitly in these reports, its genomic localization within or near such imprinted domains suggests that, as a microRNA, it may contribute to fine‐tuning or reinforcing the allele‐specific silencing through post‐transcriptional regulation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nA separate body of literature has detailed the functions of SLC22A3—also known as organic cation transporter 3 (OCT3)—in the regulation of neuromodulatory systems, drug disposition, and cellular responsiveness to hormones and stress. OCT3 is highly expressed in the brain (including key regions such as the amygdala, hippocampus, and dorsal raphe), where it mediates low‐affinity, high‐capacity uptake of monoamines (serotonin, dopamine, norepinephrine) and contributes to the maintenance of neurotransmitter tone as well as behavioral responses to stress. Similar roles extend to peripheral tissues, where OCT3 influences the absorption, tissue distribution, and pharmacologic actions of therapeutic agents such as metformin, and even mediates processes as diverse as taste perception in salivary glands and cardiovascular regulation. In many of these contexts, the quantitative and sometimes compensatory regulation of OCT3 expression (for example, in knockout or transporter variant models) highlights the importance of additional layers of gene regulation. In this regard, MIR4291 could, by targeting transcripts in this pathway‐regulating network, be posited as a modulator of OCT3 levels and/or activity, thereby influencing monoaminergic neurotransmission and drug pharmacokinetics."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "18"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional studies in developmental and immunological models further emphasize the intricate transcriptional regulation that governs non‐coding RNA expression. Research in embryonic stem cells has shown that key pluripotency factors (for example, STAT3 and Oct3/4) control the expression of target genes like Dax1, suggesting that complex regulatory circuits integrate transcription factor networks with non‐coding RNAs. Similarly, in immune cells such as basophils, early cytokine production is tightly regulated, implying that microRNAs may have broader roles in fine‐tuning rapid gene expression responses. Thus, while the direct functions of MIR4291 remain to be experimentally validated, its membership in a family of microRNAs embedded within these regulatory landscapes positions it as a likely contributor to the post‐transcriptional modulation of genes involved in imprinting, neuromodulation, metabolism, and cell differentiation."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "19"}]}, {"type": "t", "text": ""}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Frank Sleutels, Ronald Zwart, Denise P Barlow "}, {"type": "b", "children": [{"type": "t", "text": "The non-coding Air RNA is required for silencing autosomal imprinted genes."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Nature (2002)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1038/415810a"}], "href": "https://doi.org/10.1038/415810a"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "11845212"}], "href": "https://pubmed.ncbi.nlm.nih.gov/11845212"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Frank Sleutels, Grace Tjon, Thomas Ludwig, et al.
