MIR431 Gene

HGNC Family Non-coding RNAs
Name microRNA 431
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR431 is emerging as a versatile regulator in non‐malignant contexts. In models of interferon response, adipogenic differentiation and autoimmune conditions such as Graves’ disease, MIR431 expression is modulated to influence fundamental cellular processes. For example, studies have shown that in intervertebral disc degeneration MIR431 targets inhibitors of Wnt signaling, while increased expression in preeclampsia placentas and necrotizing enterocolitis has been linked to inflammatory responses via down‐regulation of transcription factors such as FOXA1. Other investigations have demonstrated that MIR431 plays roles in pulmonary function by regulating surfactant homeostasis and in chondrocyte apoptosis associated with osteoarthritis, with additional evidence pointing toward its involvement in the regulation of inflammatory processes in rheumatoid arthritis and even in tailoring host responses relevant to novel RNA therapeutics for tuberculosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the oncologic arena MIR431 exhibits context‐dependent roles that modulate tumor proliferation, invasion and metastasis. In hepatocellular carcinoma, papillary thyroid carcinoma, pancreatic cancer, and melanoma, altered levels of MIR431 have been correlated with tumor progression and patient prognosis. In lung cancer, studies have revealed that long noncoding and circular RNAs function as molecular sponges for MIR431, thereby indirectly regulating targets such as RAF1 and influencing angiogenesis and cell migration. Similar mechanisms have been observed in tongue squamous cell carcinoma, esophageal carcinoma and neuroendocrine tumors, where MIR431 affects key pathways including Ras/extracellular signal–regulated kinase signaling and epithelial–mesenchymal transition. Moreover, in glioma, ovarian, nasopharyngeal, colon, and gastric cancers the MIR431-associated regulatory axis (which includes targets like NRXN3, PBOV1, SOX9, HCAR1, PRDX1, and SLC7A11) underscores its multifaceted action in tumor growth and apoptosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the mechanistic level, MIR431 functions as a nodal regulator by interacting with various competing endogenous RNA networks across different tissue types. It directly binds to the 3′ untranslated regions of key modulators—such as SFRP1 and GSK‑3β in disc degeneration, FOXA1 in intestinal inflammation, DAB2IP in neuroendocrine tumor metastasis, and Smad4 in pulmonary surfactant regulation—to influence classical signaling cascades including Wnt, MAPK, and Ras/Erk. In addition, sponging interactions with circular and long noncoding RNAs have been shown to fine‑tune its availability and downstream effects on targets including NRXN3, SOX9, PBOV1, HCAR1, PRDX1, and SLC7A11.
