MIR4423 Gene

HGNC Family Non-coding RNAs
Name microRNA 4423
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR-4423-3p has emerged as a key regulatory molecule with distinct roles in different disease contexts. In rheumatoid arthritis (RA), miR-4423-3p is significantly downregulated in peripheral blood mononuclear cells, while its downstream target—matrix metalloproteinase 13 (MMP13)—is upregulated in the synovial tissue of affected patients. By binding directly to the 3′ untranslated region of MMP13 mRNA in RA fibroblast-like synoviocytes, overexpression of miR-4423-3p leads to decreased MMP13 expression. This modulation results in the inhibition of cell proliferation, migration, and invasion, along with the induction of cell cycle arrest and promotion of apoptosis. These findings indicate that restoring miR-4423-3p expression may serve as a novel therapeutic strategy for RA."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Conversely, during hepatitis C virus (HCV) infection, miR-4423-3p exerts a pro-viral effect by directly interacting with the mRNA of retinoic acid-inducible gene I (RIG-I). This direct binding suppresses the RIG-I-mediated interferon (IFN) response, which is crucial for the host’s antiviral defense. Consequently, the reduced activation of the IFN pathway facilitates HCV infection and replication. This contrasting role underscores the context-dependent function of miR-4423-3p, acting as a negative regulator of inflammatory and catabolic processes in RA, while simultaneously impairing innate antiviral responses in HCV-infected cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Weihong Xu, Lu Ye, Huaxiang Wu "}, {"type": "b", "children": [{"type": "t", "text": "MicroRNA-4423-3p inhibits proliferation of fibroblast-like synoviocytes by targeting matrix metalloproteinase 13 in rheumatoid arthritis."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Bioengineered (2021)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1080/21655979.2021.1988372"}], "href": "https://doi.org/10.1080/21655979.2021.1988372"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "34696684"}], "href": "https://pubmed.ncbi.nlm.nih.gov/34696684"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Xijing Qian, Bingan Wu, Chen Xu, et al.
