MIR4490 Gene

HGNC Family Non-coding RNAs
Name microRNA 4490
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In this context, previous proteomic studies have revealed that the proteasome subunit‐α type‐6 (PSMA6) protein is substantially down-regulated in nephropathic kidneys. This down-regulation was confirmed in diabetic patients using immunohistochemistry and further investigated in vitro using NRK-52E cells cultured under high glucose conditions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "In the NRK-52E cell model, although the levels of PSMA6 mRNA remained largely unaltered under high glucose conditions, PSMA6 protein expression was significantly reduced. This discrepancy pointed to a post-transcriptional regulatory mechanism. Subsequent dual luciferase reporter assays established that miR-4490 binds directly to the PSMA6 mRNA, mediating its translational suppression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "Furthermore, the inverse correlation observed between miR-4490 expression and PSMA6 protein levels, together with the modulation of PSMA6 expression upon exogenous adjustment of miR-4490, underscores the role of miR-4490 as a key regulator in diabetic nephropathy. These findings support the potential evaluation of miR-4490 as a biomarker for nephropathy in diabetic patients and provide a foundation for future therapeutic strategies targeting this microRNA."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Ying Feng, Ming-yue Jin, Dong-wei Liu, et al.
