| HGNC Family | Non-coding RNAs |
| Name | microRNA 4492 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In bladder cancer, MIR4492 functions as a tumor‐suppressive microRNA by targeting ROMO1, a protein involved in oxidative stress and cell proliferation. However, its activity is attenuated by the oncogenic long noncoding RNA LINC00319, which acts as a molecular sponge for MIR4492. The suppression of MIR4492 by LINC00319 results in elevated ROMO1 expression, thereby enhancing proliferation, migration, and invasiveness of bladder cancer cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn an inflammatory context, MIR4492 is implicated in the pathogenesis of osteoarthritis. Exposure of osteoarthritis synovial fibroblasts to IL-17 activates the MEK/ERK signaling cascade, which in turn modulates MIR4492 levels. This activation of the MEK/ERK/MIR4492 axis leads to increased production of the proinflammatory cytokine IL-18, suggesting that MIR4492 plays a role in mediating inflammatory responses within joint tissues."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nTogether, these findings highlight the versatile functions of MIR4492 across diverse biological environments. In cancer, its negative regulation by LINC00319 disrupts its tumor-suppressive effects, thereby facilitating oncogenic progression, while in osteoarthritis, its involvement in the MEK/ERK signaling axis underscores its contribution to inflammatory cytokine production. These insights into MIR4492’s dual roles open avenues for targeted therapeutic strategies in both oncologic and inflammatory disorders."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yu Yang, Fangyi Zhang, Hang Huang, et al.
