MIR450A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 450a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn colorectal carcinoma, the miR‐450a family plays an important tumor‐suppressive role. Specifically, miR‐450a-5p directly targets the stem cell transcription factor SOX2, thereby suppressing cancer stem cell properties, angiogenesis, and vasculogenic mimicry. This downregulation of SOX2 curbs the proliferation and self-renewal capacity of malignant cells, suggesting that miR‐450a functions as a critical brake on colorectal tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the heart, miR‐450a-2-3p mediates antifibrotic effects in the setting of atrial fibrillation. By directly inhibiting MAPK1 expression, miR‐450a-2-3p reduces the proliferation of cardiac fibroblasts and dampens the production of key fibrotic markers such as α-SMA, COL1, and COL3, while preserving endothelial characteristics. This modulation of MAPK1-driven signaling helps to mitigate myocardial fibrosis and improves cardiac tissue remodeling."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn breast cancer, a similar regulatory strategy is evident, as low levels of miR‐450a are associated with a more aggressive tumor phenotype. Restoration of miR‐450a expression suppresses cell proliferation, migration, and invasion by targeting CREB1 and consequently inhibiting downstream PI3K/AKT signaling and matrix metalloproteinase activities. These findings underscore the broader role of miR‐450a in restraining oncogenic signaling pathways and highlight its potential as a therapeutic target across diverse malignancies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jiaxuan Chen, Shengyuan Chen, Linghao Zhuo, et al.
