| Name | microRNA 4517 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent research into synaptic scaffolding proteins has revealed critical roles in modulating neuronal signaling and plasticity, with important implications for conditions such as traumatic brain injury (TBI). In one study, the postsynaptic scaffold protein Preso was found to regulate N‐methyl-D-aspartate receptor (NMDAR) activity and downstream nitric oxide (NO) and Ca2+ responses. Downregulation of Preso resulted in neuroprotection in TBI models, largely by dampening excitotoxic signaling mediated via its interactions with PSD-95 and downstream effectors such as neuronal nitric oxide synthase (nNOS) and cyclin-dependent kinase 5 (CDK5)."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a related investigation into neural scaffolding proteins and cognitive development, FRMPD4 was shown to participate in dendritic spine morphogenesis through its binding with PSD-95 and Homer. Disruptive mutations in FRMPD4 were linked to an X-linked intellectual disability syndrome in humans and produced spatial learning and memory deficits in mouse models, highlighting the importance of properly organized postsynaptic structures for normal synaptic function."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAlthough neither study explicitly addresses MIR4517, the context provided by these investigations suggests that microRNAs—such as MIR4517—might play an important regulatory role in fine-tuning the expression and activity of key postsynaptic scaffold proteins including Preso and FRMPD4. Such microRNA-mediated regulation could, in turn, influence NMDAR signaling pathways, calcium and NO responses, and dendritic spine dynamics, potentially impacting both neuroprotection following injury and cognitive function. This hypothesis underscores the need for further research to delineate whether MIR4517 directly or indirectly modulates these synaptic components and processes."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Peng Luo, Xin Li, Xiuquan Wu, et al.
