MIR4518 Gene

HGNC Family Non-coding RNAs
Name microRNA 4518
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Numerous investigations have demonstrated that activation of β2 integrins—such as LFA‑1 and Mac‑1—and their regulation by intracellular adaptors (e.g., Rap1, talin, kindlin‑3, ADAP) are essential for neutrophil adhesion, transmigration, dendritic cell–T‐cell interactions, and other leukocyte trafficking events. These processes are central to acute inflammation and immune surveillance."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nOther studies have extended these observations to disease models—including Alzheimer’s disease, arthritis, acute lung injury, and ischemia–reperfusion—in which modulating integrin activation and the ensuing intracellular signaling (via molecules such as PLCγ2 and specific Src family kinases) can attenuate leukocyte recruitment and tissue damage. Such data underscore the therapeutic potential of targeting integrin‐dependent adhesion pathways to interrupt harmful inflammatory cascades."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nWithin this context, microRNAs—well‐known post‑transcriptional regulators of gene expression—often fine‑tune key pathways by repressing or modulating the synthesis of proteins involved in signal transduction and cell adhesion. By analogy with other miRNAs that govern immune cell function, MIR4518 may be poised to regulate the expression or activity of pivotal integrin‐associated proteins (for example, components of the Rap1/talin/ADAP module or other cytoskeletal regulators). In this way, MIR4518 could act as a molecular rheostat to dampen excessive integrin activation and leukocyte recruitment, thereby limiting damaging inflammatory responses as observed in models of neutrophil extravasation, T‑cell trafficking, and inflammatory tissue injury."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "21"}]}, {"type": "t", "text": " Future studies are needed to determine whether MIR4518 directly targets transcripts encoding these adhesion and signaling proteins and how such regulation may contribute to the modulation of inflammatory diseases.\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Theresa T Lu, Jason G Cyster "}, {"type": "b", "children": [{"type": "t", "text": "Integrin-mediated long-term B cell retention in the splenic marginal zone."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Science (2002)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1126/science.1071632"}], "href": "https://doi.org/10.1126/science.1071632"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "12130787"}], "href": "https://pubmed.ncbi.nlm.nih.gov/12130787"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Muhammad Reza Marwali, Jose Rey-Ladino, Lisa Dreolini, et al.
