| HGNC Family | Non-coding RNAs |
| Name | microRNA 451b |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐451, including its variant miR‐451b, plays diverse roles in tumor biology across multiple cancer types. In esophageal carcinoma and renal cell carcinoma, miR‐451 functions predominantly as a tumor suppressor. In the esophageal carcinoma cell line EC9706, miR‐451 was found to inhibit cell proliferation through as‐yet‑to‑be‑elucidated molecular mechanisms, while in renal cell carcinoma, its marked downregulation is associated with higher histological grade and lymph node metastasis. Restoration of miR‐451 expression in renal carcinoma cells suppresses proliferation, migration, and invasion by directly targeting macrophage migration inhibitory factor (MIF)."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, studies in gastric cancer stem‐like cells indicate an oncogenic role for miR‐451b. Inhibition of hsa‐miR‐451b leads to reduced activities of matrix metalloproteinases MMP‑2 and MMP‑9, diminished migration, invasion, and angiogenesis, and an induction of apoptosis. These molecular and functional changes are accompanied by alterations in stemness markers and key regulators of cell survival, suggesting that miR‐451b supports the self-renewal and tumorigenic properties of these cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn glioma, miR‐451 acts as a potent inhibitor of progression and metastasis by restraining epithelial-mesenchymal transition (EMT).
