MIR4530 Gene

HGNC Family Non-coding RNAs
Name microRNA 4530
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent findings in human umbilical vein endothelial cells have demonstrated that overexpression of a novel microRNA—designated miR‐14530 in this study—leads to a marked suppression of cell proliferation and an enhancement of apoptosis. This activity appears to be mediated via the upregulation of RASA1, a gene that, upon increased expression, modulates both the extracellular signal‐regulated kinase (ERK)/mitogen‑activated protein kinase (MAPK) and phosphoinositide‑3‑kinase (PI3K)/AKT signaling pathways. Although the precise mechanism by which miR‐14530 enhances RASA1 protein levels remains to be fully elucidated, these observations underscore its potential role as a negative regulator of cellular growth."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn parallel, studies on human gliomas have revealed that miR‐4530 is significantly down‑regulated in clinical tissues and glioma cell lines, such as U251 and T98G. Upon forced overexpression, miR‐4530 exerts potent tumor‑suppressive effects, including the inhibition of cell proliferation, colony formation, migration, and invasion, as well as the promotion of apoptotic cell death. These anti‑tumor activities are achieved through the direct targeting of RTEL1, a gene whose elevated expression counteracts the suppressive influence of miR‑4530 on malignant phenotypes, a finding further supported by in vivo xenograft models."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, the data from these studies highlight the critical role of these microRNAs in controlling cellular homeostasis via the regulation of key target genes. While one investigation demonstrates that miR‐14530 can inhibit endothelial cell growth through modulation of RASA1 and its downstream ERK/MAPK and PI3K/AKT pathways, another study establishes that miR‐4530 functions as a tumor suppressor in gliomas by targeting RTEL1. These complementary findings suggest that modulation of such microRNAs could represent a promising therapeutic strategy in conditions characterized by aberrant cell proliferation."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Li Jing, Hong Li, Tao Zhang, et al.
