MIR4537 Gene

HGNC Family Non-coding RNAs
Name microRNA 4537
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have established the critical roles of netrin‐1 receptors—particularly the UNC5 family—in mediating apoptosis, neural development, and cell migration. For example, UNC5H1 interaction with NRAGE is required for netrin‐1‐independent apoptosis, and distinct expression patterns of Unc5 receptors have been observed during mouse embryogenesis, including in the central nervous system and developing vasculature. Additionally, post‑translational modifications such as C‑mannosylation have been shown to be crucial for proper trafficking of UNC5A from the endoplasmic reticulum to the cell surface, while genetic ablation studies underscore UNC5A’s involvement in neuronal apoptosis and its regulation as a p53 target in various cancers. Moreover, structural insights into Unc5D’s assembly with glypican‑3 provide a broader context for the conserved mechanisms governing cell guidance and migration."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast to these receptor‐centered studies, research in gastric cancer has identified miR-4537 as a key regulatory microRNA that functions as a tumor suppressor. Specifically, miR-4537 is downregulated in gastric cancer tissues and cell lines, and its restoration inhibits cell proliferation while promoting apoptosis. Mechanistic investigations revealed that miR-4537 directly targets the zinc finger protein ZNF587 by binding to its 3′ untranslated region, leading to suppressed ZNF587 expression. Importantly, overexpression of ZNF587 was able to partially reverse the pro-apoptotic and anti-proliferative effects of miR-4537, and elevated miR-4537 levels were found to improve the radiosensitivity of gastric cancer cells. These findings highlight miR-4537 as a promising therapeutic candidate for enhancing radiotherapy efficacy in gastric cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Megan E Williams, Phyllis Strickland, Ken Watanabe, et al.
