| HGNC Family | Non-coding RNAs |
| Name | microRNA 454 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐454, particularly its –3p species, displays potent oncogenic functions in a variety of human cancers. In several studies of breast, prostate, gastric, colorectal, cervical, and nasopharyngeal carcinomas, for example, overexpression of miR‐454 was shown to promote cell proliferation, migration, invasion, epithelial–mesenchymal transition and chemoresistance by directly inhibiting key tumor‐suppressive genes (such as RPRD1A, CHD5, CYLD, TRIM3, and NDRG2) and concomitantly activating oncogenic pathways including Wnt/β‐catenin signaling. In hepatocellular carcinoma, elevated miR‐454 levels in tumor‐initiating cells further contributed to self‐renewal and sorafenib resistance, positioning this miRNA as a potential prognostic indicator as well as a therapeutic target in these settings."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "12"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, a number of investigations in glioblastoma, non–small cell lung cancer, oral squamous cell carcinoma, anaplastic thyroid carcinoma, neuroblastoma, and even colon cancer have demonstrated that miR‐454 can function as a tumor suppressor. In these studies, ectopic restoration of miR‐454 expression impaired cell cycle progression and reduced cell proliferation, migration, and invasion by targeting oncogenic mediators including PDK1, NFATc2, CPEB1, and NR3C2. In studies of pre‐eclampsia, decreased miR‐454 levels in placental tissues were linked to impaired trophoblast proliferation and invasion—phenomena mediated in part by targets such as ALK7 and EPHB4—thereby contributing to disease pathogenesis. Similarly, in models of heart failure and ischemic stroke, miR‐454 exerts cardioprotective and neuroprotective effects by modulating apoptotic pathways and oxidative stress (through regulators like NEDD4‐2/TrkA and NOX4). In vascular settings, including sickle cell disease and hereditary hemorrhagic telangiectasia, miR‐454 negatively regulates vasoactive and profibrotic factors (ET‐1 and PAI‐1) and supports proper angiogenic function, while its downregulation is associated with bone invasion in pituitary adenomas and altered disease activity in multiple sclerosis. Notably, a potential role in acute myeloid leukemia has also been suggested, although its precise contributions in this context remain to be fully elucidated."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "21", "end_ref": "29"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Hong-Liang Liang, Ai-Ping Hu, Sen-Lin Li, et al.
