| HGNC Family | Non-coding RNAs |
| Name | microRNA 4638 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In castration resistant prostate cancer (CRPC), miR-4638-5p is found to be significantly underexpressed compared to androgen dependent prostate cancer. Functional analyses have demonstrated that restoration of miR-4638-5p suppresses androgen-independent growth both in vitro and in vivo by directly targeting Kidins220. This targeting results in decreased activity of the PI3K/AKT pathway as well as a reduction in VEGF‐mediated angiogenesis, suggesting that loss of miR-4638-5p may contribute to the development of castration resistance and increased tumor aggressiveness."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n\nSeparately, investigations in metastatic triple-negative breast cancer have identified that TGF‑β1 downregulates miR-4638-3p expression. This downregulation relieves the inhibitory control on ATF3, leading to its increased expression and subsequent activation of pathways that promote tumor cell proliferation, invasion, and bone metastasis. Thus, MIR4638 (via its −3p form) emerges as a critical regulator of metastasis by modulating ATF3 levels and its downstream effectors, indicating its potential as both a biomarker and therapeutic target in metastatic breast cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yang Wang, Ning Shao, Xueying Mao, et al.
