| Name | microRNA 4649 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐4649‑3p plays a tumor‐suppressive role in nasopharyngeal carcinoma (NPC) by directly targeting key regulatory molecules. In NPC cell lines, elevated levels of miR‑4649‑3p are associated with reduced expression of the tyrosine phosphatase SHP‑1. Functional experiments using miR‑4649‑3p mimics and inhibitors have demonstrated that up‐regulation of miR‑4649‑3p suppresses NPC cell proliferation through down‐regulation of SHP‑1, an effect that is reversible upon exogenous SHP‑1 expression. This inverse correlation underlines a direct regulatory relationship whereby miR‑4649‑3p limits tumor cell growth by targeting SHP‑1."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a complementary study, in silico RNA crosstalk analyses revealed that miR‑4649‑3p can bind both to the long non‐coding RNA LINC02570 and to SREBF1 mRNA. In advanced-stage NPC, overexpression of LINC02570 appears to sequester miR‑4649‑3p, thereby diminishing its availability to suppress SREBF1. This molecular “sponge” effect leads to the up-regulation of SREBP1 and its downstream target, fatty acid synthase (FASN), promoting lipid metabolic changes that favor tumor progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xiaofen Pan, Gang Peng, Sha Liu, et al.
