MIR4674 Gene

HGNC Family Non-coding RNAs
Name microRNA 4674
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

In patients with prostate cancer, elevated levels of miR-4674 have been observed in the metastatic form compared to non-metastatic disease, and its candidate target genes (including JUNB, KMT2A, and XPO6) suggest a potential role in promoting tumor progression and metastasis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, investigations focused on tissue repair and angiogenesis following myocardial injury have demonstrated that miR-4674 is significantly downregulated in affected patients. Functionally, overexpression of this human-specific microRNA in endothelial cells markedly impairs key angiogenic processes such as proliferation, migration, network tube formation, and spheroid sprouting. Mechanistic studies indicate that miR-4674 exerts its influence by modulating VEGF-stimulated p38 MAPK signaling through direct targeting of interleukin 1 receptor-associated kinase 1 (Irak1) and Bicd cargo adaptor 2 (Bicd2), while its inhibition yields pro-angiogenic effects."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional research into inflammatory conditions such as psoriasis—where macrophage activation and mitochondrial dynamics have been highlighted as key factors—did not identify a role for miR-4674. Although the pathogenesis of psoriasis involves complex signaling cascades, miR-4674 was not implicated in these processes, suggesting that its regulatory functions may be limited to pathways involved in cancer metastasis and vascular remodeling rather than in broader inflammatory responses."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "E N Knyazev, T R Samatov, K A Fomicheva, et al.
