MIR4742 Gene

HGNC Family Non-coding RNAs
Name microRNA 4742
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have identified miR-4742-5p as an important regulatory molecule in cancer progression. In non‐small cell lung cancer (NSCLC), miR-4742-5p is significantly upregulated and functions to target and downregulate TACC1, a protein involved in cellular organization and growth. This targeting enhances tumor cell invasion and cisplatin resistance, while reducing apoptosis. Notably, the long noncoding RNA LINC01140 acts as a competing endogenous RNA (ceRNA) by sponging miR-4742-5p, thereby indirectly upregulating TACC1 and mitigating the aggressive behavior of NSCLC cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n \n \nDisruptions in miR-4742-5p expression are also implicated in gastric cancer (GC) progression. In GC cell lines, increased levels of miR-4742-5p correlate with enhanced proliferation, migration, and invasion. Mechanistic investigations have shown that miR-4742-5p induces epithelial-mesenchymal transition (EMT) by downregulating adhesion molecules such as Zo-1 and E-cadherin, while upregulating mesenchymal markers like vimentin and N-cadherin. Furthermore, Rab43 was identified as a potential direct target, influencing the activation of critical oncogenic pathways including AKT and NF-κB."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n \n \nCollectively, these findings underscore the dual roles of miR-4742-5p in different cancer types, functioning as an oncogenic factor through distinct molecular mechanisms. In NSCLC, its regulation via LINC01140 and targeting of TACC1 modulates chemoresistance and cell survival, whereas in GC, miR-4742-5p promotes EMT and metastatic potential through its effects on cellular adhesion and key signaling pathways. This emerging microRNA may represent a promising therapeutic target in tailored cancer treatment strategies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yanyun Wang, Man Li, Lin Zhang, et al.
