| HGNC Family | Non-coding RNAs |
| Name | microRNA 484 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐484 (miR-484) has emerged as a key regulator in several aspects of cancer biology and therapy. In ovarian and renal cell carcinoma, for example, miR-484 expression is intricately linked with chemoresistance and tumor vascular remodeling. In ovarian cancer, altered miR-484 levels predict resistance to chemotherapy through modulation of the VEGFB/VEGFR2 signaling axis and can be efficiently delivered via RGD‐modified exosomes to normalize tumor vessels and enhance chemosensitivity. Similarly, in metastatic renal cell carcinoma, lower tissue miR-484 levels are associated with prolonged time to progression, underscoring its critical role in tuning drug response."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn diverse cancer types, miR-484 displays context-dependent functions that affect tumor progression. In hepatocellular carcinoma, miR-484 is sequestered by circADAMTS13, thereby exerting tumor suppressor effects; in colorectal cancer, long noncoding RNAs such as ZFAS1 and pgm5-as1 sponge miR-484, promoting cell proliferation and invasion. In non-small-cell lung cancer, high miR-484 levels drive migration and proliferation via direct repression of Apaf-1, leading to reduced apoptotic signaling. Moreover, in gastric cancer, miR-484 acts as a tumor suppressor by targeting CCL-18 and concomitantly impeding PI3K/AKT signaling, while in breast cancer models, its modulation influences key regulators such as cytidine deaminase and KLF4, thereby affecting both chemoresistance and cancer stem cell-like properties."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumor growth and chemoresistance, miR-484 also contributes to the regulation of cellular homeostasis in non-malignant contexts. In the reproductive system, elevated miR-484 in granulosa cells from patients with diminished ovarian reserve impairs cell viability and promotes apoptosis by directly targeting YAP1 and disrupting mitochondrial function. In endothelial cells, shear stress–dependent regulation of miR-484 processing via nucleolin (modulated by AMPK) influences the expression of vasoprotective factors such as KLF2 and eNOS, thereby impacting vascular homeostasis. Similarly, exosomal miR-484 derived from bone marrow mesenchymal stem cells has been shown to suppress pancreatic cancer cell proliferation and migration by disrupting mitochondrial metabolism through inhibition of the Wnt/MAPK signaling pathway."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "11", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Andrea Vecchione, Barbara Belletti, Francesca Lovat, et al.
