| HGNC Family | Non-coding RNAs |
| Name | microRNA 485 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐485 – expressed as both miR‐485-3p and miR‐485-5p – has emerged as a powerful tumor suppressor in a wide spectrum of cancers. In prostate, breast, hepatocellular, lung (both non‐small cell and small cell), bladder, melanoma, osteosarcoma, and colorectal cancers, miR‐485 is frequently downregulated and its loss is correlated with enhanced cell proliferation, migration, invasion, epithelial–mesenchymal transition (EMT) and resistance to chemotherapeutic agents. Mechanistically, miR‐485 directly targets multiple oncogenic regulators such as IGF2BP2, FZD7, survivin, MUC1, NTRK3, stanniocalcin 2, WBP2, TPX2, and key signaling mediators (AKT1, HSP90), thereby inhibiting pathways like Wnt/β‐catenin, PI3K/AKT and MAPK/ERK. Moreover, several studies have shown that competing endogenous RNA (ceRNA) networks—including long noncoding RNAs (e.g. NEAT1, DSCR8, SNHG6, SNHG11) and circular RNAs (e.g. circPTCH1, circ‐KRT6C)—can “sponge” miR‐485, modulating its availability and further influencing tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncogenesis, miR‐485 plays versatile roles in non‐malignant contexts. In viral infections, for instance, miR‐485 modulates the innate immune response by targeting retinoic acid-inducible gene I (RIG-I) to prevent excessive antiviral signaling and, under high viral loads, directly binding viral RNA polymerase gene PB1 to limit influenza virus replication. During iron deficiency, miR‐485-3p is upregulated to repress ferroportin expression, contributing to the fine-tuning of iron homeostasis. Its regulatory reach also extends to immunoinflammatory and degenerative diseases: in cutaneous lupus erythematosus, enhanced miR‐485-3p levels in leukocytes lead to T-cell activation and fibroblast-driven fibrosis, while in diabetic nephropathy, miR‐485 suppresses mesangial cell proliferation and pro-inflammatory cytokine production via downregulation of NOX5. Moreover, in Alzheimer’s disease, elevated miR‐485-3p correlates with pathological hallmarks such as amyloid-β accumulation and tau truncation, with antisense inhibition ameliorating these toxic effects; emerging data also suggest a potential role for miR‐485-3p in osteoarthritis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "22", "end_ref": "28"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its direct actions on gene targets, miR‐485 is itself regulated by complex ceRNA networks that have significant therapeutic implications. In hematologic settings, for example, reduced miR‐485-3p levels lead to upregulation of transcriptional mediators such as NF-YB and consequent alterations in topoisomerase IIα expression, affecting drug sensitivity. Similarly, in lung cancers and other disorders, long noncoding RNAs—such as SNHG6 and NEAT1—act as molecular sponges for miR‐485, thereby modulating downstream targets like VPS45, BSG, and AIM2; these interactions not only influence processes like EMT and cell proliferation but also impact systemic immune balances, as observed in preeclampsia.
