| HGNC Family | Non-coding RNAs |
| Name | microRNA 487b |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In tobacco‐induced lung carcinogenesis, for example, miR‐487b is epigenetically repressed, resulting in the upregulation of targets involved in oncogenic signaling (such as SUZ12, BMI1, WNT5A, MYC, and KRAS), thereby contributing to malignant transformation (a tumor‐suppressive function when active)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn several tumor types—including pediatric gliomas, colorectal and colon cancers, hepatocellular carcinoma, and neuroblastoma—miR‐487b is found at reduced levels, and its re‐expression inhibits key malignant behaviors such as proliferation, migration, invasion, and colony formation. In these contexts, miR‐487b exerts tumor‐suppressive effects by directly targeting oncogenes or molecules that mediate aggressive cancer phenotypes (for instance, GRM3, KRAS, LRP6, MYC, and SUZ12), and its expression level can serve as a prognostic marker that improves risk stratification."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn other settings the function of miR‐487b appears more nuanced. In non–small cell lung cancer models of acquired temozolomide resistance, higher levels of the miR‐487b–5p isoform are associated with enhanced cell proliferation, migration, and chemoresistance—effects that are partly mediated by repression of molecules such as LAMP2. Similarly, in thyroid cancer, miR‐487 (together with miR‐154) can collaboratively modulate RHOA signaling. Notably, studies in osteosarcoma have reported seemingly contradictory roles: one investigation shows that diminished miR‐487b–3p levels correlate with increased metastasis and chemoresistance via targeting ALDH1A3, whereas another indicates that miR‐487b is overexpressed and promotes malignant behaviors through TRAK2 regulation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumorigenesis, miR‐487b also participates in non–tumorigenic processes. Moreover, in models of inflammatory injury—such as lipopolysaccharide‐induced microglial activation relevant to spinal cord injury—and in studies probing its involvement in allergic rhinitis via the IL‑33–ST2 axis, miR‐487b demonstrates the capacity to modulate inflammation and cellular responses."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Sichuan Xi, Hong Xu, Jigui Shan, et al.
