| HGNC Family | Non-coding RNAs |
| Name | microRNA 493 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‐493 functions in a context‐dependent manner across a spectrum of malignancies. In several cancer types including bladder, colon, hepatocellular, melanoma, and non‐small cell lung cancers, miR‐493 exerts tumor‐suppressive effects by attenuating cell proliferation, migration, invasion, and metastasis. For example, in bladder cancer miR‐493 downregulates oncogenes such as FZD4 and RhoC to inhibit cell motility, while in colon cancer it suppresses liver metastasis by targeting key mediators including IGF1R and MKK7. Similarly, in hepatocellular carcinoma and melanoma, restoration of miR‐493 reduces tumor growth by inhibiting GP73, MYCN, VAMP2, and IRS4, and its targeting of ITGB1 in non‐small cell lung cancer is associated with improved patient outcomes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nThe expression and activity of miR‐493 are frequently modulated by epigenetic mechanisms and by competitive endogenous RNA networks. In hepatocellular carcinoma, miR‐493‐5p silencing via hypermethylation contributes to tumor progression, whereas in various cancers ceRNA molecules—such as lncRNAs (e.g., CCAT2 in cervical cancer and GAS6‑AS2 in breast cancer) and circular RNAs (e.g., circPRKDC in gastric cancer and LINC00324 in esophageal squamous cell carcinoma)—sequester miR‐493‐5p.
