MIR5000 Gene

Name microRNA 5000
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nStudies have revealed that miR‑5000‑3p is upregulated in specific tumor cell subpopulations. In colon cancer, for instance, this microRNA is markedly elevated in the side population of cells—an aggressive subset implicated in multidrug resistance. This upregulation suggests that miR‑5000‑3p may serve as a potential biomarker for identifying drug‐resistant cancer stem-like cells, thereby offering a novel target for overcoming resistance mechanisms in advanced colon cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition, investigations in laryngocarcinoma have elucidated an oncogenic role for miR‑5000‑3p. Here, the long noncoding RNA MIR22HG, which normally acts as a tumor suppressor, functions by sponging miR‑5000‑3p, thereby restraining its activity. When MIR22HG is downregulated—partly due to transcriptional inhibition by E2F6—miR‑5000‑3p levels rise, leading to the repression of the downstream target FBXW7, a gene crucial for inhibiting cell proliferation and migration. Consequently, elevated miR‑5000‑3p contributes to enhanced tumor cell proliferation and migration in laryngocarcinoma, underscoring its potential as an oncogenic driver in this context."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Zhong-Sheng Xia, Lin Wang, Tao Yu, et al.
