| Name | microRNA 5006 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough none of the presented abstracts mention MIR5006 directly, they collectively delineate a network of stress‐responsive, translational regulatory processes principally mediated by the heme‐regulated eIF2α kinase (HRI) and other eIF2α kinases. In conditions such as arsenite exposure, viral infection, and oxidative/metabolic stress, HRI plays a central role by phosphorylating eIF2α to attenuate global protein synthesis while selectively promoting translation of key stress response factors (e.g., ATF4). In doing so, HRI supports cellular survival by enabling stress granule formation and by initiating a cytoprotective unfolded protein response that coordinates innate immune signaling – functions that one might speculate are modulated by regulatory RNAs such as MIR5006 to fine‐tune these pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nOther studies emphasize HRI’s indispensable role in erythroid cells and macrophages, where it senses heme deficiency to balance globin synthesis and to regulate iron homeostasis. HRI activation in these cells adjusts translation through eIF2α phosphorylation, modulates ATF4‐dependent transcriptional programs, and even influences mTORC1 signaling, thereby coordinating responses during iron deficiency, anemia, or inflammatory stress. Such tightly controlled translational responses may be further impacted by microRNAs such as MIR5006, which could conceivably act to modulate HRI expression or the translation of its downstream effectors in these settings."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nA further collection of findings outlines HRI’s contribution to proteostasis and specialized cellular functions including the clearance of cytosolic protein aggregates, regulation of innate immunity during bacterial infection, and modulation of synaptic plasticity. Notably, HRI‐dependent signaling supports the degradation of misfolded proteins via autophagy, restricts toxic oligomer formation in neurodegenerative contexts, and enhances NMDA receptor function by upregulating specific subunits. Although the direct involvement of MIR5006 is not described, one might hypothesize that MIR5006 could participate in these regulatory circuits by targeting transcripts of HRI or its effectors, thereby influencing the balance of protein synthesis and stress adaptation across diverse cellular settings."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Edward McEwen, Nancy Kedersha, Benbo Song, et al.
