MIR5009 Gene

Name microRNA 5009
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough none of the abstracts directly mention MIR5009, the collective studies provide an extensive framework on cyclin D1 regulation—a pathway that MIR5009 is now emerging to modulate. In normal development and cell cycle progression, cyclin D1 governs the transition from G1 to S phase, and its precise expression is essential for processes such as neural progenitor differentiation, adipocyte differentiation, and β‐cell expansion. For instance, work in mouse embryogenesis shows that alterations in cell cycle length through cyclin D1–dependent mechanisms are necessary for the switch from proliferation to neurogenesis, while studies in fibroblasts reveal that cyclin D1 nuclear dynamics regulate cell cycle exit and apoptosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn many pathologic settings—ranging from inflammation to oncogenesis—cyclin D1 becomes deregulated, and its overexpression or altered subcellular localization can lead to unchecked proliferation. Studies highlight that aberrant cyclin D1 not only accelerates tumor initiation but also contributes to processes such as chromosomal instability, metabolic deregulation, and resistance to apoptosis. In several tumor models, cyclin D1 overexpression is linked to enhanced signaling through pathways such as Ras/MAPK, NF‑κB, and insulin/Akt, leading to a feedback network that reinforces its oncogenic expression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "24"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nEmerging evidence also underscores the complexity of cyclin D1 regulation at both transcriptional and post‐transcriptional levels. Advanced studies describe how cyclin D1 functions beyond merely catalyzing cell cycle progression; it also modulates transcriptional programs, chromatin structure, and even mitochondrial function. These multifaceted roles are controlled by its interplay with various signaling cascades and interacting proteins, including factors that regulate internal ribosome entry and mRNA translation. Furthermore, contexts such as stem cell proliferation in the hippocampus and aberrant growth factor responses in cancer models reveal that fine‐tuning of cyclin D1 levels is imperative for maintaining cellular homeostasis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "25", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nGiven this intricate regulatory landscape, MIR5009 is likely to function as a pivotal post‐transcriptional regulator that fine‑tunes cyclin D1 expression. By targeting cyclin D1 mRNA—either through modulating its translation, stability, or degradation—MIR5009 could influence critical cellular decisions such as proliferation versus differentiation, tumor suppression versus oncogenesis, and metabolic balance versus dysregulation. In effect, MIR5009 may serve as an “integrator” that links external signaling cues (via pathways like Ras/MAPK, NF‑κB, and insulin/Akt) to the internal execution of cell cycle programs, making it a promising candidate for therapeutic intervention in conditions where cyclin D1 is aberrantly expressed.\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Karin Barnouin, Marlène L Dubuisson, Emma S Child, et al.
