| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent investigations have revealed that miR-5047 plays a critical role in modulating osteosarcoma cell behavior through its interaction with long noncoding RNA WAC‐AS1. Specifically, miR-5047 normally targets the mRNA of the transcription factor SOX2, serving to suppress its expression and thereby constrain cell proliferation and stemness. However, in osteosarcoma, WAC‐AS1 is highly expressed and acts as a competing endogenous RNA, binding to miR-5047 and sequestering it. This interaction diminishes miR-5047’s inhibitory effect on SOX2, allowing SOX2 levels to rise and promoting enhanced tumor cell proliferation and metastatic potential."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFurthermore, the stimulation of SOX2 not only drives oncogenic cell stemness and metastasis but also reinforces a positive feedback mechanism. SOX2 directly binds to the promoter region of WAC‐AS1, further boosting its transcription and perpetuating the cycle of miR-5047 inhibition. Consequently, the WAC‐AS1/miR-5047/SOX2 axis emerges as a pivotal regulatory pathway in osteosarcoma malignancy, highlighting miR-5047’s function as a tumor suppressor that is subverted during disease progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Zhining Yang, Zhaoyong Liu, Weiqing Lu, et al.
