MIR506 Gene

HGNC Family Non-coding RNAs
Name microRNA 506
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐506 (miR-506) has emerged as a multifaceted regulator in cancer biology, exerting potent tumor‐suppressive effects in a broad range of malignancies. In ovarian, breast, cervical, gastric, colorectal, pancreatic, prostate, thyroid, esophageal, lung, neuroblastoma, osteosarcoma and other cancers, miR-506 predominantly inhibits cell proliferation, migration, invasion, and the epithelial-to-mesenchymal transition (EMT), thereby curbing metastasis and enhancing apoptosis. In ovarian cancer, for example, miR-506 augments E-cadherin expression and suppresses mesenchymal markers by directly targeting EMT drivers such as SNAI2, while in cervical and breast cancers it downregulates oncogenic transcription factors like Gli3 and IQGAP1, respectively. These findings are supported by studies in diverse cancers that demonstrate miR-506’s role in counteracting tumor aggressiveness and chemoresistance by impeding EMT characteristics and related signaling pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the molecular level, miR-506 orchestrates its tumor-suppressive functions by directly targeting a range of oncogenes and signaling mediators. It represses key drivers of cellular transformation such as SNAI2, SPHK1, CDK4/CDK6, Gli3, EZH2, IQGAP1, YAP1, ABCC4, CREB1, JAG1, SHH, PIM3, and even MYCN in neuroblastoma, thereby disrupting proliferative, survival, and angiogenic pathways. Furthermore, miR-506’s expression is subject to epigenetic regulation and can be modulated by interactions with long non-coding RNAs and circular RNAs (for instance, NEAT1, circUBAP2, or KCNQ1OT1 that act as sponges), which in turn impact its availability to target downstream effectors. Notably, these regulatory interactions extend to influencing autophagy-related cell death—as observed via modulation of the STAT3-BCL2-BECN1 axis in pancreatic ductal adenocarcinoma and by regulating autophagic flux in osteosarcoma—thereby sensitizing tumors to chemotherapeutic agents."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "11", "end_ref": "23"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumor cell intrinsic processes, miR-506 also influences the tumor microenvironment and non-malignant tissues. In cholangiocytes, its upregulation is linked to impaired bile secretion and cholestatic injury, while in the cardiovascular context, miR-506 (in cooperation with miR-124) confers protection against oxidative stress by targeting transcription factors such as KLF4 and KLF5. Circulating levels of miR-506 have been evaluated as potential diagnostic and prognostic biomarkers in esophageal and colorectal cancers, and in select cases, elevated plasma levels correlate with poorer outcomes, highlighting its context-dependent behavior. In addition, miR-506 affects drug sensitivity and immune escape mechanisms—as noted in studies of non-small cell lung cancer and hepatocellular carcinoma—thus representing a promising candidate for combinatorial therapeutic strategies."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "24", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Ani Manichaikul, Qian Wang, Yu Lena Shi, et al.
