MIR512-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 512-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR512-1, giving rise to mature miR‐512-5p and miR‐512-3p species, has emerged as an important regulator of cell fate by modulating apoptosis and proliferation in diverse disease contexts. In gastric cancer cells, epigenetic activation of MIR512-1 (miR‑512‑5p) promotes apoptosis through the suppression of the anti‐apoptotic protein Mcl‑1, while in head and neck squamous cell carcinoma, miR‑512‑5p directly targets hTERT to reduce telomerase activity and tumor cell proliferation. In Alzheimer disease brain regions with tau pathology, reduced MIR512 expression is accompanied by increased levels of its targets, including c‐FLIP and Mcl‑1, suggesting that altered MIR512-1 levels may disturb the balance between pro- and anti‑apoptotic signals."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistically, MIR512-1 exerts its functions by targeting and downregulating multiple key transcripts. In hepatocellular carcinoma cells, miR‑512‑3p reduces c‑FLIP expression, thereby enhancing taxol-induced apoptosis, while in non‑small cell lung cancer, regulation of a miR‑512‑5p/p21 axis modulates cell proliferation, migration, and invasion. Moreover, in osimertinib-resistant lung adenocarcinoma, MIR512-1 is sponged by circular RNAs resulting in the derepression of IGF1R, contributing to drug resistance, and in both atherosclerosis models and retinoblastoma cells, miR‑512‑3p targets the endoplasmic reticulum stress regulator XBP‑1, affecting apoptosis and autophagy."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "8"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these studies illustrate that MIR512-1 acts as a multifaceted tumor suppressor and a modulator of cellular stress responses. Its ability to target anti-apoptotic and proliferation-associated factors such as Mcl‑1, c‑FLIP, hTERT, IGF1R, and XBP‑1 positions MIR512-1 as a promising therapeutic target across a range of conditions from malignancies—including gastric, hepatocellular, head and neck, lung, and retinal cancers—to neurodegenerative and vascular disorders.
