MIR513A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 513a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nCurrent studies have revealed that miR‐513a variants serve as important post‐transcriptional regulators in diverse biological contexts. In human granulosa cells, miR‐513a‐3p negatively regulates luteinizing hormone/chorionic gonadotropin receptor (LHCGR) expression by binding to multiple sites within its 3'UTR, thereby modulating receptor levels and impacting reproductive signaling processes. In parallel, in renal cell carcinoma, elevated levels of circMYLK promote tumorigenesis by sponging miR‐513a‐5p, which normally would down‐regulate VEGFC; the sequestration of miR‐513a‐5p leads to increased VEGFC expression, contributing to tumor growth and metastasis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nOther investigations extend these findings to additional cancer types. In gastric cancer, circBFAR acts as a sponge for miR‐513a‐3p, thereby relieving its suppressive effect on hexokinase 2 (HK2) expression. The resulting upregulation of HK2 not only enhances cancer cell proliferation but also increases glycolytic activity. Similarly, in nephroblastoma, the long non‐coding RNA DLEU1 exerts oncogenic effects by modulating the miR‐513a‐5p/RANBP2 axis—its interaction with miR‐513a‐5p contributes to decreased apoptosis and supports tumor cell survival."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn a different cellular setting, research in human myeloid leukemia cells (K‑562) has demonstrated that circRNA.0007127 controls apoptosis via the miR‐513a‐5p/CASP8 pathway. Silencing of this circRNA leads to reduced caspase‑8 activation and decreased apoptotic rates, while overexpression of miR‐513a‐5p diminishes CASP8 levels, further underscoring the central role of miR‐513a in regulating cell survival mechanisms. Together, these studies illustrate that miR‐513a variants, modulated by various competing endogenous RNAs, are pivotal in governing key processes such as proliferation, apoptosis, and metabolic reprogramming across distinct disease contexts."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "B Troppmann, N Kossack, V Nordhoff, et al.
